磷酸化蛋白组
在酸性条件下,TiO2表面带正电,可与磷酸化肽段产生静电作用而达到富集效果。经TiO2富集后的多肽产物再进行液相色谱质谱(LC-MS)分析,即可获得磷酸化肽段的序列信息和磷酸键的位置信息。
TiO2与iTRAQ技术的结合,可实现磷酸化蛋白组的鉴定和定量比较。在TiO2富集之前先对所有肽段进行iTRAQ标记,利用LC-MS对比样品中各磷酸化肽段iTRAQ信号强度,即可获得磷酸化蛋白组的定量信息。
